On my manifold the heater core hose runs to driver side and temp sensor on passenger side by tstat.
All pics I see that’s reversed?
Does it matter?

Got a picture?
Ultimately it probably doesn’t matter, but there might still be some variation in flow for the heater core, and a variation on temperature reading. But probably not much.

However, the fitting for the heater is supposed to be an elbow, with a siphon tube extending down into the coolant.
This, or a lack thereof, could make the difference.

Given the huge difference in size between the elbow and sensor threads, I wonder how they are installed.
Which model intake is it?

The bosses have different thread sizes.
As Paul mentioned the elbow's siphon tube needs clearance in the manifold, and I dont think there is enough depth on driver side.
After coolant exits intake manifold it needs to pass thru heater temp control valve (in proper direction) before going to heater core.
Heater hose on drivers side may not play nice with carb linkage or distributor, depending on how it is routed.

The real short answer, it doesn't matter. The is a common passage where water from both heads joins together for a single thermostat.

Do you have the distributor installed yet? That driver's side port can cause clearance issues if trying to use a heater hose there. Just bulkier than a compact little temp sensor. And that also plays into what distributor as well.
